The present work describes the characterization of the sulfur-oxidizing enzyme in Acidihiobacillus ferrooxidans. Oxidation of elemental sulfur by a cell-free preparation of Acidithiobacillus ferrooxidans was first described by Silver & Lundgren (1968). Like the Acidithiobacillus ferrooxidans cell-free system described by Suzuki & Silver (1965), this system required glutathione (GSH) for its activity and oxidized elemental sulfur to sulfite.Sulfur oxygenase was a cytoplasmatic enzyme with pH optimum 7.4.The isoelectric point of the enzyme was 6.0. The relative molecular weight of the enzyme under native condition was assessed as 500 000. Glutathione - polysulfide could be used as a substrate by the enzyme and reduced glutathione could be replaced by sulfide.
