Purine derivates - separation and characterization by selected methods
Original language description
Purine bases represent the basic constituents of DNA, their analysis is useful in the diagnosis of several diseases and metabolite disorders. The related bases, such as 6-aminopurine (6-AP, 2-aminopurine (2-AP) and 2,6-diaminopurine (2,6-DAP) were studies by spectroscopic and RP-HPLC techniques. Studied APs can be separated based on their spectral properties. Elution order related to polarity of molecule, in MF MetOH/30mM phosphate buffer (15:85), was following: 2,6-DAP, 2-AP, 6-AP. The phosphate concentration is not crucial. Experimental pKa values of model APs were calculated from corrected capacity factor dependence of APs on pH. The basicity increasing in order 2-AP<6-AP<2,6-DAP. Studies of basic structures will be used for nucleosides and nucleotides study, where 2-AP and 2,6-DAP can be used as fluorescent labels.
