    Auxin and cytokinin (CK) are principal regulators of the developmental fate of pluripotent plant cells. However, the molecular nature of their interaction during de novo organogenesis remains unclear. We used the hypocotyl explants-based in vitro systemto study mechanisms underlying de novo organogenesis. We show that auxin, but not CK, is able to induce organogenic response. CK modulates auxin-induced organogenesis (AIO) via regulation of intercellular auxin distribution. The AIO is accompanied with endogenous CKs production and tissue-specific activation of CK signalling. CK reduces auxin efflux from cultured tobacco cells and regulates the expression of PIN auxin efflux carriers in hypocotyl explants. We analyzed PINs expression in roots of 6-day-old seedlings and found that PIN2 and PIN4 are downregulated in Pro35S:AtCKX2(3) lines. We propose a model in which auxin acts as a trigger of the organogenic processes, whose output is modulated by the endogenously produced CKs.
