In the virological practice the preconcentration, purification and subsequently the determination of the purity and the concentration of the viruses from the cultural medium and/or from the real sample are required. The conventional techniques used todayare equipment demanding, time-consuming and labourious. In this study user friendly virus purification from the biological sample and the determination of the resulting viral sample purity by the capillary isoelectric focusing with UV detection are introduced. The Equine and Swine influenza viruses from the infected allantoic fluid from specific pathogen free embryonated chicken eggs were precipitaded by PEG 6000 and sodium chloride. The precipitated viruses were sedimented already at the accelerationabout 14 000 g. The impurities with different densities were removed on the sucrose gradients.
