The establishment of iterative patterns in body shape and architecture is a common characteristic in many organisms. The underlying developmental mechanisms in plants are poorly understood. In plants, new organs are periodically initiated in a regular pattern called phyllotaxis. Self-organization of phyllotaxis is governed by the formation of morphogen-based inhibitory fields. Interaction of the morphogens auxin and cytokinin was found to be fundamental for organ development. Using RNA-Seq, we identified several cytokinin metabolic enzymes (LOGs) to be potentially regulated by auxin. Additionally, we inspected single mutants to test genetically the contribution of particular enzymes towards meristem patterning. To explore in more detail functional significance, we use synthetic biology tools to understand quantitatively the response to auxin and analyze the response dynamics. This approach will allow the analysis of auxin-cytokinin cross-regulation in the dynamics of shoot apical meristem patterning.
