    The hybrids of F1 generations usually experience heterosis for fitness-related traits (including the resistance to parasites), whilst post-F1 generations, due to Dobzhansky–Muller genetic incompatibilities, express numerous disadvantageous traits (including susceptibility to parasites). Genetic disruption especially in post-F1 generations of hybrids may also result from the broken system of cyto-nuclear coadaptation. Maternal backcrosses (parents with the same mtDNA) and paternal backcrosses (parents with different mtDNA) have the same nuclear genetic compositions, but differ in cytoplasmic genetic elements, affecting their viability and survival. We used spring viremia of carp virus (SVCV), associated with common carp, to examine heterosis versus breakdown in hybrids of the cyprinid species - common carp and gibel carp. Common carp was highly susceptible to SVCV, whilst gibel carp was less susceptible. F1 generations exhibited heterosis advantage, with the expression of SVCV similar to that in less susceptible gibel carp. Paternal backcrosses and F2 generations, burdened by high cyto-nuclear incompatibility, tended to exhibit SVCV dynamics similar to that of highly susceptible common carp. The patterns of hybrid heterosis and hybrid breakdown were also revealed by measuring dynamics of anti-SVCV specific antibodies in blood serum. We employed high-throughput RNA sequencing to generate transcriptomic profiles of head kidney. Hybrid heterosis for F1 generations was mostly reflected by similarity in the down-regulation of differentially expressed genes corresponding to the viral infection disease. High hybrid breakdown was documented throughout the transcriptomic analysis for paternal backcross generations and F2 generation of hybrids, confirming the broken system of cyto-nuclear coadaptation in those post-F1 generations. This evidence may support the role of intrinsic selection acting against inter-specific hybridization in wild-living freshwater fish.
