    A method for the measurement of ascorbic acid (AA) using high-performance liquid chromatography with ultraviolet detection and possibilities of protein precipitation with regard to stability and recovery are described. The aim of this work was to find out the most suitable precipitant reagent for the determination of AA in consideration of stability and recovery. The effectiveness of various protein precipitants was tested as follows. To plasma and AA solution (50.0 uM), the following different precipitant reagents were added: metaphosphoric acid (5%), perchloric acid (1.0 M), trichloroacetic acid (10%), sulfosalicylic acid (10%), acetonitrile with hydrochloric acid, acetonitrile with acetic acid, methanol with hydrochloric acid, and ethanol with hydrochloric acid. The only two protein precipitants that led to satisfactory recoveries of AA were metaphosphoric and perchloric acid. Stability of AA samples for analysis was investigated over 10 hours.     V této práci je popsána metoda pro stanovení kyseliny askorbové (AA) pomocí vysoceúčinné kapalinové chromatografie s UV detekcí a možnosti precipitace proteinů s ohledem na stabilitu a výtěžnost. Cílem této práce bylo nalézt nejvhodnější precipitační činidlo pro stanovení AA ve smyslu stability a výtěžnosti. Testovali jsme různá precipitační činidla. K plazmě a roztoku AA (50 umol/l) jsme přidávali tato precipitační činidla: kyselinu metafosforečnou (5%), kyselinu chloristou (1,0 mol/l), kyselinu trichloroctovou (10%), kyselinu sulfosalicylovou (10%) acetonitril okyselený kyselinou chlorovodíkovou, acetonitril okyselený kyselinou octovou, methanol okyselený kyselinou chlorovodíkovou a ethanol okyselený kyselinou chlorovodíkovou. Pouze s dvěmi precipitačními činidly jsme dosáhli dostatečné výtěžnosti, a to s kyselinou metafosforečnou a kyselinou chloristou. Stabilita AA ve vzorcích byla sledována po dobu 10-ti hodin.
