Electrochemical Analysis of DNA: When, How and Why to Use Electroactive Labels and Indicators
Original language description
DNA possesses intrinsic electrochemical activity due to the presence of electrochemically reducible or oxidizable nucleobases and exhibits characteristic, structure-sensitive adsorption/desorption behavior at mercury or amalgam electrodes. Thus, label-free electrochemical DNA sensing is in principle possible, and especially in connection with mercury-based or carbon electrodes the intrinsic DNA electroactivity has been utilized to detect DNA damage. In other applications, such as sequence-specific DNA sensing, typing of single-nucleotide polymorphisms (SNP) etc., utilization of electroactive labels has appeared advantageous. We present several examples of using redox-active or enzymatic DNA tags in the detection of DNA damage, DNA hybridization and minisequencing of the SNP.
