Polygalacturonase immobilized by adsorption on carrier from PET bottles
Original language description
Polygalacturonase is the part of pectolytic complex which has wide industrial application. By the immobilization of enzyme the improvement of its industrial application properties can be achieved. Commercially used pectolytic complex Rohament P was irreversibly adsorbed on carrier prepared from polyethyleneterephtalate (PET) bottles, what is a new patent of Brno University of Technology. Immobilization of the enzyme resulted in lowering of polygacturonase activity, the measure of which depended on the amount of the enzyme fixed on the carrier. The highest relative activity (96%) had the preparation containing 10 micrograms of the enzyme per 1 g of the carrier. The activity of all preparations was lowered when compared with that of the soluble enzyme. It was dependent on the amount of the enzyme adsorbed on the carrrier. No residual activity of non-adsorbed enzyme was found in any of the solution after a 4 h incubation. Activity of the free and immobilized polygalacturonase was determin
