Original language description
The reported system is based on the temperature measurement system that was developed in the first stage of the sub-project. It serves for contactless, non-destructive temperature measurement of vitrified samples in electron microscope (EM). The spatial resolution is not limited by a conventional diffractive limit but in principle should allow temperature estimates of the sample (cell) surface on the scale of tens of nanometers. The sensing principle is based on cathodoluminescence (CL) of colloidal quantum dots deposited over a sample of interest. The functional sample is based on a modified CL detector that allows direct measurement of CL. The major progress is represented by the detailed calibration and optimization of the device parameters.

An integral part of any temperature measurement technique is precision and accuracy evaluation and calibration. The measurement technique based on cathodoluminescence (CL) of quantum dots, which was developed in the earlier stage of the project, had to be calibrated with the aid of full-range spectrometry. For this, an optical fiber was introduced into the chamber of an electron microscope and the in-situ CL spectra were evaluated with respect to a calibration measurement of photoluminescence in an external cryostat. The obtained results were used to create a calibrated look-up table which could be used to translate the RGB signal output of the CL detector. The sensitivity and accuracy of the functional sample is supposed to be improved by utilization of narrow- band RGB filters.
Economical parameters
The device will form a basis for future temperature evaluation within dual beam systems for preparation of cryo-samples.
