    The Effect of Focal Plane on the SERS performance of a Gold-Epoxy Nanocomposite

    In this work, the effect of proper focal depth on the obtained surface-enhanced Raman spectroscopy (SERS) signal on a nanostructured surface has been studied. For this, ellipsoidal gold nanoparticles arranged in a hexagonal array on top of an epoxy substrate were used. The gold nanoparticles were functionalized with DNA molecules via thiol-chemistry. By using a confocal microscopy setup and a pinhole, the Raman spectrum of the sample was recorded with 100 nm resolution in the normal (Z) direction in the ±15 μm range. The results confirm that Raman peaks characteristic to DNA molecules originate only from a narrow ~100 nm region of the substrate (in the normal direction), which proves that by properly focusing the laser beam on the substrate and filtering out of focus light with a pinhole, the SERS enhancement can be optimized. © 2021 IEEE.
