    Angiopoietin-like protein 4: development, analytical characterization, and clinical testing of a new ELISA

    We developed and evaluated the sandwich ELISA method for the quantitative determination of human Angplt4 in serum. We conducted a study on individuals with metabolit syndrome (MS) or familiar hypercholesterolemia and 30 healthy probands and measured blood pressure, waist circumference (WC), Angplt4 serum levels, serum cholesterol, triglycerides (TG), HDL-cholesterol, LDL-cholesterol, insulin, glucose, A-FABP and calculated BMI and QUICKI index. Furthermore, we tested 115 individuals with MS Probands with MS did not differ in Angplt4 values in healthy individuals. Angplt4 values correlated with the HDL-cholesterol, glucose, TG and number or characters of metabolic syndrome. No significant correlation was found between serum Angplt4 and BMI, WC or QUICKI. We found that Angplt4 was not an independent marker for metabolic syndrome. The study supports the role of Angplt4 as a new class of lipid metabolism modulator.

    Vyvinuli a otestovali jsme sandwichovou ELISA metodu pro kvantitativní stanovení lidského Angiopoietin-like protein 4 (Anglpt 4) v séru. Provedli jsme studii s jedinci, kteří trpí metabolickým syndromem (MS) či familiární hypercholesterolémií a 30 zdravými jedinci. U všech subjektů byly měřeny hodnoty krevního tlaku, obvodu pasu, v séru stanoveny koncentrace Anglpt 4, cholesterolu, triglyceridů (TG), HDL-cholesterolu, LDL- cholesterolu, insulínu, glukózy, A-FABP a vypočteny BMI a QUICKI index. Navíc bylo otestováno 115 jedinců s MS. Hodnoty Anglp4 u jedinců s MS se nelišily od hodnot u zdravých jedinců. Angplt4 koreloval s HDL cholesterolem, glukózou, TG a počtem komponent metabolického syndromu. Nebyl zjištěna korelace mezi Angpl4 a BMI, obvodem pasuči QUICKI. Bylo zjištěno, že Angplt4 není nezávislým markerem metabolického syndromu. Studie podporuje úlohu Angplt4 jako nově objeveného modulátora metabolismu lipidů.
