Contribution (poster) in 44th ESNA Confrence in Brno 1 - 5 September, 2015, p. 65. The aim of this study was quantification of native plant growth regulators in barley kernels. In caryopses samples of 12 selected barley varieties were determined cytokinins content, ACC (1-aminocyclopropane-1-carboxylic acid, the precursor of ethylene) and ABA (abscisic acid) using of extraction and purification procedures (ion exchange chromatography and reverse phase chromatography), using combined instrumental (GC and HPLC) and immunoassay techniques (ELISA, RIA) with high detection sensitivity (pg / 50 l).HPLC separations allowed the quantification of individual cytokinins - concretly 10-bioactive cytokinins (trans-zeatin, dihydrozeatin, isopentenyladenine, benzyladenine, meta-topolin and their ribosides). They were found in most of isoprenoid cytokinins trans-zeatin (tZ) and dihydrozeatin (DHZ) and its riboside (DHZR), aromatic benzyladenine (BA) and its riboside (BAR) and meta-topolinribosid (mTR). Its quantity aromatic cytokinins at samples exceeded isoprenoid cytokinins. Representation of the various forms of cytokinins in selected varieties was relatively variable - the highest content (tens of ng / g fresh weight) were found in the varieties Bojos, Francin, Laudis 550 Vendela. Higher ACC contents were determined in the kernel of KM 2454 (floury), Bojos, Petrus and Vendela. In varieties Nudimelanocrithon was detected three to four time higher levelof this substances. ACC low levels were quantified in KM 1057 and Tocada. Abscisic acid in kernels of varieties have exceeded 50 ng / g of fresh weight.
