Agrobacterium ? mediated transformation of faba bean (Vicia faba L.) to induce resistance to PEMV and PSbMV viruses.
Original language description
Pea seed-borne mosaic (PSbMV) and Pea enation mosaic viruses (PEMV) belong to frequent viral pathogens causing losses in several grain legumes including faba bean. These losses might be prevented by growth of resistant varieties and transgenic technologies might provide the mean for the crop protection when naturally occurring resistances are not available. Knowledge of in vitro regeneration of faba bean originated from meristems of embryogenic axis was utilized. We have used faba bean (Vicia faba L.) cultivars Merlin, Merkur, Mistral and Agrobacterium-mediated genetic transformation to provide resistances to both viruses. Transformation was performed with A. tumefaciens EHA105 strain harbouring the T-DNA constructs of coat viral protein (cp) fragmentsof either PEMV and/or PSbMV in sense-antisense orientation under the control of 35S CaMV promoter to induce RNA mediated silencing. High score (up to 80 %) and positive transfected tissues (more than 50 % of total area, measurements by i
