Utilizing SILAC quantification approach and high resolution mass spectrometry we monitor the differences in membrane proteome of Francisella tularensis subsp. tularensis and Francisella tularensis subsp. holarctica. Even these subspecies differ by 85% ongenome level we were able successfully quantify over 500 hundreds proteins. Based on quantification study we selected: eleven proteins that are uniquely expressed in Francisella tularensis subsp. tularensis or Francisella tularensis subsp. holarctica and 32 proteins differing in expression level. Subsequently we expressed these proteins in E. coli and used them to produce rabbit polyclonal antibodies. We confirmed our SILAC results and antibody specificity using combination of peptide mass fingerprinting and western blots.
