    Identification of interspecific barriers in the genus Lotus

    The aim of our research study is determinate barriers of crossability after interspecific hybridization. An interspecific hybridization program was undertaken involving the nonshattering species Lotus conimbricensis (2n = 12), L. uliginosus (2n = 12, 2n=14), L. ornithopodioides (2n = 14, and they could be crossed with Lotus corniculatus (2n = 24. To study pre-fertilization barriers, callose in growing pollen tubes were stained by aniline blue. To investigate the role of post-fertilization barriers, hybrid embryo viability was traced by clearing treatment of immature seed using chloral hydrate after modification. In Lotus the optimal time of fixation was 24 hours and length of clearing by chloral hydrate was 24 hours. This method was successfully applied for ovule clearing 24 days after pollination.

    Cílem našeho studia byla determinace bariér křížitelnosti po mezidruhové hybridizaci u rodu Lotus. Hybridizační program zahrnoval nepukavé druhy Lotus conimbricensis (2n = 12), L. uliginosus (2n = 12, 4n = 24), L. ornithopodioides (2n = 14), které byly kříženy s kulturním druhem Lotus corniculatus (2n = 24). Bariéry křížitelnosti můžeme rozdělit na prezygotické a postzygotické. Pro studium prezygotických bariér byla zvolena metodika barvení kalózy v pylových láčkách anilinovou modří. Postzygotické bariéry zahrnující defekty ve vývoji embrya byly studovány pomocí metodiky projasňování rostlinných pletiv pomocí chloralhydrátu. Optimální čas fixace byl 24h a projasňování v chloralhydrátu probíhalo po dobu 24dní.
