    Affinity of extracellular phosphatases for ELF97 phosphate in aquatic environments

    We indirectly studied the affinity of extracellular phosphatases for ELF97 phosphate (ELFP) in different aquatic environments through its ability to inhibit the hydrolysis of 4-methylumbelliferyl phosphate. Values of inhibition constants, Ki, which correspond to the concentrations necessary for half saturation of phosphatases by ELFP,were lowest in the oligotrophic Mediterranean Sea.We found higher values in oligo- to mesotrophic acidified lakes, in a eutrophic reservoir and in a pure culture of the marine bacterium Alteromonas infernus. Our experiments indicated that the assay ELFP concentration in the endogeneous phosphatase detection kit (Molecular Probes) used would ensure >99% saturation of extracellular phosphatases in marine environments and >90% saturation in the studied fresh waters.

    Studovali jsme afinitu extracelulárních fosfatáz pro substrát ELF97 fosfát (ELFP) v různých akvatických ekosystémech nepřímou metodou založenou na inhibici hydrolýzy 4-metylumbeliferyl fosfátu. Hdnoty inhibiční konstanty, Ki, které odpovídají koncentracipotřebné pro 50-% saturaci enzymu ELFP, byly nejnižší v oligotrofní oblasti Středozemního moře, vyšší v acidifikovaných jezerech, eutrofní přehradě a v kultuře bakterie Alteromonas infernus. Naše pokusy ukazují že koncetrace ELFP v kitu na měření endogenních fosfatáz (Molecular Probes) zajišťuje >99% saturaci extracelulárních fosfatáz v moři a >90% saturaci ve sladkých vodách.
