    Cyclodextrin glucanotransferase synthesis by semicontinuous cultivation of magnetic biocatalysts from cells of Bacillus circulans ATCC 21783

    Cells of Bacillus circulans ATCC 21783 were used as a model for preparing of magnetic biocatalysts applied for cyclodextrin glucanotransferase (CGTase) synthesis. The cell immobilization was carried out with four types of magnetic nano- and microparticles. The highest CGTase production was achieved after 96 h of semicontinuous process using silanized magnetite covalently bound on the cell surface. The specific enzyme activity was 8.4-fold higher compared to that of free cells. Magnetic nanoparticles linked on the cell walls by ionic interactions were partially released during the cultivation, while the covalent bond between the activated magnetite and the cells was very stable.

    Buňky Bacillus circulans ATCC 21783 byly použity jako model pro přípravu magnetických biokatalyzátorů pro syntézu cyklodextrin glukanotransferasy (CGTasa). Imobilizace buněk byla provedena se 4 typy magnetických nano- a mikročástic. Nejvyšší produkce CGTasy bylo dosaženo po 96h semikontinuální kultivaci buněk kovalentně vázaných na silanizovaný magnetit. Specifická enzymová aktivita byla 8,4krát vyšší než u neimobilizovaných buněk. Magnetické nanočástice spojené s buněčnou stěnou iontovými interakcemi byly v průběhu kultivace částečně uvolněny, zatímco kovalentní vazba mezi aktivovaným magnetitem a buňkou byla stabilní.
