    Juvenile hormone (JH) prevents metamorphosis of insect larvae to pupae and adults, however, a molecular mechanism of JH action remains unknown. By using RNA interference in the red flour beetle Tribolium castaneum, we discovered that the transcription factor Broad-Complex (BR-C) coordinates the transition between larval, pupal and adult insect stages during metamorphosis. We further showed that JH regulates metamorphosis by altering the expression of the BR-C gene through the Methoprene-tolerant (Met) protein. This study for the first time implicated Met in JH-dependent regulation of a gene that is required for metamorphosis.

    Juvenilní hormon (JH) zabraňuje přeměně hmyzích larev na kukly a dospělce, avšak molekulární mechanismus působení JH zůstává neznámý. S využitím interference RNA u brouka Tribolium castaneum jsme zjistili, že transkripční faktor Broad-Complex (BR-C) koordinuje přeměnu mezi larvou, kuklou a dospělým stadiem hmyzu během metamorfosy. Dále jsme ukázali, že JH řídí metamorfosu tím, že mění expresi genu BR-C prostřednictvím proteinu Methoprene-tolerant (Met). Tato práce poprvé prokázala funkci Met v JH-závislé regulaci genu, nezbytného pro metamorfosu.
