Lyme borreliosis, known as Lyme disease in the USA, is the most common vector-borne disease in the temperate regions of the northern Hemisphere. It is caused by spirochetes of the Borrelia burgdorferi sensu lato complex and is transmitted by the bite of Ixodes ticks.1, 2 More than 120 000 cases are reported annually by health-care providers, laboratories, and health authorities to official national surveillance systems in Europe (2005–20)3 and more than 33 000 cases are reported in the USA (2008–21).4 However, based on data from insurance claims in the USA, up to 476 000 patients with clinically suspected Lyme borreliosis are diagnosed and treated each year in the USA,5 indicating a potential annual national incidence rate of around 143 cases per 100 000. According to these estimates, the risk of contracting Lyme borreliosis by the age of 77 years (the average life expectancy in the USA) is around 10%, which means that one in 10 people will be infected at least once in their lifetime. Although preventive measures such as the use of repellents, regular self-examinations, and the immediate removal of ticks (within 24 h of attachment) can reduce the risk of infection,6 the number of Lyme borreliosis cases continues to rise, underscoring the urgent need for an effective vaccine.
