Physiological production of hydrogen sulfide is involved in porcine oocytes maturing in vitro
Original language description
Oocyte meiotic maturation is indispensable process of female gametogenesis. Hereby matured oocyte is predestined for fertilisation and subsequent embryonic development. The running of meiotic maturation is a result of activity of key factors – M-phase/Maturation Promoting Factor (MPF) and Mitogen Activated Protein Kinase(MAPK). Based on present knowledge, we hypothesise physiological action of hydrogen sulfide produced by CBS, CTH and 3-MPST, hydrogen sulfide releasing enzymes, during in vitro oocyte maturation.Oocytes were in vitro matured in modified M199 culture medium for 48 hrs in 39°C and 5% CO2. Consequently, parthenogenic activation of Na2S-treated oocytes by calcium ionophore A23185 was performed for evaluation of developmental competence. Our results demostrate the presence of mRNA CBS, CTH and 3-MPST CBS, CTH and 3-MPST themselves in porcine oocytes. The inhibition of present enzymes causes the deceleration of meiotic maturation and failure in achievement of fertilizing ability in a regula
