The effect of Cannabis extracts on the adhesion of lactic acid bacteria in in vitro intestinal model
Original language description
Cannabis is mainly known for its psychotropic effects due to its THC and CBD content. However, it contains a variety of other biologically active substances that have an impact on the gut microbiota. Since cannabis can be included in human nutrition, it has the potential to positively influence the adhesion of lactic acid bacteria to intestinal epithelial cells. The influence of cannabis extracts from different maturity periods was investigated. Tested were samples harvested in the tenth and fourteenth weeks of vegetation at 50, 10, and 3 µg/ml concentrations. L. gasseri, L. reuteri and L. plantarum strains were selected to determine their ability to modulate adhesion. The human cell lines Caco-2 and HT29 were used as a model, and Fluorescein isothiocyanate was used to determine adhesion, with which the individual bacterial strains were stained. The bacteria and the test samples were left on the cell lines for 1.5 days after staining. Subsequently, the cells were washed, and fluorescence was measured on a spectrophotometer at 495 nm. The average adhesion values ranged from 45 to 127%. These especially were L. gasseri on the Caco-2 line with cannabis extract harvested at week ten at a concentration of 10 µg/ml, L. reuteri on HT29 with both cannabis extracts at the same concentration of 50 µg/ml and L. plantarum on both lines with cannabis from week ten at the identical concentration of 10 µg/ml. Higher adhesion occurred with the cannabis harvested at ten weeks than with the 14-week-old cannabis, which is due to the different maturity stages, where the phenolic and terpene content decreases with increasing plant age. It is, therefore, clear that cannabis and its extracts could be used as an adhesion stimulator for lactic acid bacteria with an optimum harvest time at ten weeks of maturity.
