Natural forests in the Šumava mountains region were changed significantly by the applied forest management in the past few centuries. At the present time the domination of cultivated and artificially made forests is clearly visible in the region. However, some remains of supposedly original and natural stands can be found in the Šumava National Park (the first generation after the virgin forest). The classical methods of research which use phenotype characteristics or methods based on accessible historical data are not sufficient neither in determining of genetic variability nor the origin of certain population. Only modern scientific procedures can secure necessary identification data, particularly methods based on molecular biology and genetics. These methods work with so called identification markers which are under full genetic control. Our laboratory successfully mapped the genetic structure of more than 60 populations of Norway spruce (Picea abies (L.) KARST.) across the whole Cz
