Metabolic Fingerprinting Strategy to Reveal Food Fraud: Investigation of Markers for the Detection of Extra Virgin Olive Oil Adulteration with Soft-Deodorized Olive Oils

Extra virgin olive oil (EVOO) is a high-value commodity, thus might be subject of various fraudulent practices. This study was focused on a challenging authentication issue: addition of a lower grade, soft-deodorized olive oil (SDOO) to EVOO. For this type of olive oil fraud, widely accepted, reliable markers were not available, thus this fraudulent practice could be hardly recognized by official control laboratories. In the first step of the study, authentic EVOOs, SDOOs, and their admixtures were extracted by aqueous methanol. The obtained polar fractions were then analyzed by metabolic fingerprinting approach employing ultra-high performance liquid chromatography coupled to hybrid quadrupole time-of-flight high-resolution tandem mass spectrometry (UHPLC-HRMS/MS). SCIEX TripleTOF 6600 was selected because of versatility in combining high-resolution MS data collection, fast acquisition rates and high sensitivity. Subsequent chemometric evaluation suggested several m/z ions that might be characteristic for SDOOs. In the second phase, the marker ions were employed for target analysis by UHPLC-MS/MS with triple quadrupole linear ion trap mass analyzer, SCIEX QTRAP 6500, which enabled achieving lower detection limits of selected ions in EVOO/SDOO admixtures. The best markers suitable for the detection of SDOO addition were tentatively identified as methyl ester of hydroxylated octadecenoic acid and ester derivative of oleic acid. These marker compounds can be monitored in EVOO samples using a relatively simple LC-MS/MS-based method. As a result, the initially complex analytical challenge has been solved using the described HRMS-based metabolic fingerprinting strategy that, in the end, generated an analytical solution suitable for routine food testing.
