Original language description
Fourier transform (FT) Raman spectroscopy with the near infrared excitation enabled many of the limitations imposed by sample fluorescence and photoreactivity to be overcome. The method allow to fulfill the conditions of non-destructive analysis as expressed by Schrader et al.. Hence, the application of FT Raman spectroscopy and microscopy are widespread from quite simple inorganic materials to very complex biological samples. The development of non-destructive methodology is especially important in thecase of analysis of biological samples either in vitro or in vivo. The aim of this contribution is to demonstrate on a few examples the key role of optimization of both experimental setup and data evaluation procedure.
