    Chromatographic behavior of a series of N-methylated tetra and octapeptides on a reversed-phase sorbent was studied considering the information obtained on the compounds by NMR. The modified tetrapeptides were derived from GFFY-NH2, GFFF-NH2 and GFFH-NH2primary structures by N-methylation at various peptide bond positions. Similarly, the N-methylated octapeptides were based on TPK(Pac)T C-terminally elongated forms of GFFY and GFFF. It was found that many studied N-methylated peptides provide broad peaks as a consequence of cis/trans isomerism of the R1CO-N(CH3)R2 peptide bond. The extent of the peak spreading was found to be strongly dependent on the following important factors: the nature of the surrounding amino acid residues, the location of the modified peptide bond within the peptide chain, temperature and mobile phase flow-rate.     Bylo studováno chromatografické chování N-methylovaných tetra- a okatapeptidů v souvislosti s informacemi získanými NMR. Modifikované tetrapeptidy byly odvozeny od GFFY-NH2, GFFF-NH2 a GFFH-NH2 methylací na různých místech řetězce. Podobně, okatapeptidybyly odvozeny od TPK(Pac)T C-terminálně prodloužených tetrapeptidů GFFY a GFFF. Ukázalo se, že mnoho N-methylovaných peptidů poskytovalo široké chromatografické píky v důsledku cis/trans isomerie R1CO-N(CH3)R2 peptidové vazby. Rozšiřování píků závisí nanásledujících faktorech: charakter sousedících aminokyselin, poloha modifikace na řetězci, teplota, rychlost mobilní fáze. Všechny vlivy byly podrobně zkoumány. Rychlou chromatografií na 20-30 um RP sorbentu se podařilo téměř zcela separovat konformery GF(NMe)FY-NH2.
