Radio-frequency impedance based biomass measurements represent a relatively accurate and reliable method for on-line quantification of viable cellular biomass. For the living biomass concentration determination, a linear correlation between biomass concentration and capacitance is generally assumed. However, this assumption is not always valid as e.g. the presence of intracellular lipid inclusions may violate the assumption of linear correlation. Therefore, the main objective of this study was to examine the potential of the sensor for on-line in-situ monitoring of intracellular biopolymer content (PHA) in bacterial cells. It was experimentally confirmed that the loss of linearity between dry cell weight and capacitance takes place also in the case ofthe Pseudomonas putida culture. The resulting nonlinear behaviour can be used for the design of a novel biopolymer content estimator based on the capacitance and offgas composition measurements.
