    The main goal of regenerative medicine is to treat or replace organs, their parts or cells that have been damaged by age, trauma or pathological changes. For such purpose scientists work on the development of modern methods utilizing stem cells. These cells have a unique ability to differentiate in the body into any conventional cell, helping to regenerate damaged tissues. The use of stem cells for clinical applications requires high quality and quantity of cells. Traditional methods for maintenance and expansion of cells rely on culturing techniques using plastic culture plates. The problem arises in the separation of the grown cells from the substrate surface. Conventionally, cells are recovered by mechanical cell scraping or enzymatically induced cell harvesting. Both methods can be very harsh and may lead to cells with impaired cell functionality, which is a major obstacle for approval of cell therapy. The utilization of thermoresponsive poly(N-isopropylacrylamide) (pNIPAAm) surfaces for cell harvesting can be an excellent alternative to traditional methods, as the cell detachment is mediated exclusively by temperature. The study aims the preparation of the pNIPAAm modified polydimethylsiloxane substrates and characterization of their thermoresponsive behavior for future utilization in temperature-triggered cell harvesting.
