Amino acid residues in the cytoplasmic domain of the Mason-Pfizer monkey virus glycoprotein critical for its incorporation into virions

  • Original language description

The contribution of individual amino acids in the cytoplasmic domain Env of Mason-Pfizer monkey virus (M-PMV) to the process of glycoprotein incorporation was analysed using alanine-scanning mutations. The glycoprotein biosynthesis and function, as wellas virus infectivity have been examined. Mutation of valine 20 and histidine 21 inhibits viral protease mediated cleavage of the cytoplasmic domain. An isoleucine at position 18 and a leucine at position 25 each mediate the process of incorporation and efficient release of virions.

Aminokyseliny cytoplasmatické domény glykoproteinu opičího Mason-Pfizerova viru důležité pro jeho inkorporaci do virionů

  • Czech description

Vliv jednotlivých aminokyselin cytoplasmatické domény Env opičího Mason pfizerova viru byla analyzována pomocí alaninové skenovací mutageneze. Sythesa a funkce glykoproteinů a infectivita mutantů Env byly srovnány s údaji pro divoký typ viru. Mutace valinu 20 a histidinu 21 inhibovala štěpení cytoplasmatické domény Env virovou proteasou. Dále bylo zjištěno, že isoleucin v pozici 18 a leucin v pozici 25 zprostředkovává proces inkorporace a účinné uvolnění virionů.
