    The role of S-S bridge in retroviral protease function and virion maturation

    Retroviral proteases are active in a form of homodimer. We found out that protease from Mason-Pfizer monkey virus contains an intramonomeric disulfide bridge that stabilizes the dimerization interface and increases proteolytic activity in vitro. Disruption of this disulfide in the context of the whole virus (construct M-PMVPRC7A/C106A) did not have any influence on proteolytic cleavage of viral polyproteins during maturation process. However, the long term infectivity of M-PMVPRC7A/C106A was noticeablycompromised when compared to the wild type of virus.

    Retrovirové proteasy jsou aktivní ve formě homodimeru. Zjistili jsme, že proteasa Mason-Pfizerova opičího viru obsahuje intrapodjednotkový disulfidový můstek, který stabilizuje dimerizační rozhraní a zvyšuje proteolytickou aktivitu enzymu. Záměna obou cysteinových zbytků tvořících tento můstek za alaniny neměla žádný podstatný vliv na proteolytickou aktivitu uvnitř zrající virové částice, prokazatelně ovšem snížila dlouhodobou infektivitu konstruktu M-PMVPRC7A/C106A ve srovnání s virem divokého typu.
