    We herein report on immunodiagnosis of fasciolosis, and significant protection of sheep against challenge infection with Fasciola gigantica following immunization with peptides based on the H-Asp110-Lys-Ile-Asp-Trp-Arg-Glu-Ser-Gly-Tyr-Val-Thr-Glu-Val123-OH (Fas14p) sequence of F. gigantica cathepsin L-cysteine proteinase. This sequence was synthesized as 1) N .alpha. acetylated (FasAc14p), 2) bearing at the aminoterminus an N .alpha.-acetylated cystein (FasAcCys14p), and 3) conjugated in four copies tosequential oligopeptide carrier Ac-[Lys-Aib-Gly]4-OH (Ac-SOC4) through an amide bond (Ac-[Lys(Fas14p)-Aib-Gly]4-OH). The peptides were synthesized manually by SPPS on a Wang resin using the Fmoc/tBu strategy. All compounds were tested as immunodiagnostics for sheep fasciolosis. The data together indicated that the peptides might be of considerable use for discriminating between early and late, and low and high burden, sheep infection with F. gigantica.

    Práce popisuje imunodiagnostiku fasciolosy a signifikantní protekci ovcí vůči infekci parazitem Fasciola gigantica po imunizaci peptidy založenými na sekvenci H-Asp110-Lys-Ile-Asp-Trp-Arg-Glu-Ser-Gly-Tyr-Val-Thr-Glu-Val123-OH (Fas14p) z F. gigantica cathepsin L-cysteine proteinasy. Tato sekvence byla syntetizována jako 1) N alfa acetylovaná (FasAc14p), 2) nesoucí na aminokonci N alfa-acetylovaný cystein (FasAcCys14p), a 3) konjugovaná ve čtyřech kopiích na sekvenční oligopeptidový nosič Ac-[Lys-Aib-Gly]4-OH (Ac-SOC4) pomocí amidové vazby (Ac-[Lys(Fas14p)-Aib-Gly]4-OH). Peptidy byly syntetizovány manuálně metodou na pevné fázi (SPPS) na Wangově pryskyřici pomocí Fmoc/tBu strategie. Všechny látky byly testovány jako imunodiagnostika ovčí fasciolosy. Získané výsledky ukazují, že peptidy mohou mít značný význam pro rozlišení mezi časnou a pozdní, a nízkou a vysokou zátěží u infekce ovcí s F. gigantica.
