New approach to 9-[2-hydroxy-3-(phosphonomethoxy)propyl]adenine (iso-HPMPA) is described and a series of iso-HPMPA derivatives substituted at the C-8 position of the purine ring was prepared. Synthesis of the key alkylating agent was developed starting from allyl alcohol.
