Original language description
This chapter describes recent developments and applications of capillary electrophoresis coupled with mass spectrometry (CE-MS) in peptidomics. Peptidomics is introduced as a relatively new research field dealing with comprehensive qualitative and quantitative analysis of peptides in biological samples. In peptidomic, similarly as in proteomic studies, CE-MS represents a valuable alternative and/or complementary approach to the most often used techniques, different modes of liquid chromatography hyphenated with MS detection (LC-MS). Combination of these two approaches significantly enhances amounts of peptides found and identified in complex biological samples. The advantage of CE-MS is that it requires extremely low amounts of samples (usually less than LC-MS) and provides fast highly efficient separation and highly sensitive detection of peptides in complex matrices. In addition to the basic characteristics of CE-MS, sample preparation techniques commonly applied in peptidomics before CE separation and possibilities for peptide identification and quantification by CE-MS methods are discussed in this chapter. Moreover, the relevant recent application of CE-MS in several peptidomic studies are presented. The important studies dealing with the discovery or determination of peptide biomarkers for various diseases in body fluids will be shown and the peptidomic research focused on finding and identification of peptides with special biological activities in foods is demonstrated.
