GleNAc-Terminated Glycodendrimers Are Up-Taken by and Modulate NK cells Expressing NKR-P1 Receptor

  • Original language description

N-acetyl-glucosamine coated PAMAM tetradendrimers were labelled with either fluorescein or rhodamine to follow their cell targeting and fate. Cells with high content of granuli and expressing both NK cell receptors and products (IFN-gamma, granzyme B, perforin) showed membrane binding and clathrin-dependent intracellular uptake of the dendrimers. The presence of NKR-P1 receptor resulted critical and first results demonstrated its binding by the glycodendrimer with intragranular colocalization at the confocal microscopy.

GlcNAc glykodendrimery jsou inkorporovány NK buňkami nesoucími NKR-P1 receptor a modulují jejich funkci

  • Czech description

Tetradendriméry PAMAM -N-acetyl-glukosamin (PAMAM-GN4) značené buď fluoresceinem nebo rhodaminem byly sledovány z hlediska buněčné lokalizace. Buňky s vysokým počtem granulí obsahující IFN-gamma, granzym B, perforin a exprimující receptory buněk NK, bylyschopny vázat a a pohlcovat dendriméry za účasti kladrinu. Konfokální mikroskopii získané výsledky poukazují na vnitrobuněčnou kolokalizaci dendrimérů s NKR-P1 ke granulím v závislosti na vazbě k lektinovému receptoru NKR-P1.
