    Effect of Phosphate on the Expression of Protein-Ser/Thr Kinase Pkg2 in Streptomyces granaticolor

    A time-correlated expression of eukaryotic-like Ser/Thr protein kinase Pkg2 of Streptomyces granaticolor was investigated by RT-PCR and by transcription fusion experiments. In a complex medium the activity of pkg2 promotor was constant during the life cycle. Direct RNA analysis proved the presence of corresponding pkg2 transcript. S1 nuclease protection analysis of the transcription initiation site showed that pkg2 gene is a leaderless mRNA. Under phosphate starvation the promotor activity was detectable merely in the early exponential phase. Under these conditions turning off of the pkg2 promotor and cessation of pkg2 transcript level coincided with the start of granaticin production

    Exprese Pkg2, Ser/Thr proteinkinasy eukaryotního typu, u Streptomyces granaticolor byla studována pomocí RT-PCR a transkripční fúze s reportérovým genem. V komplexním médiu byla aktivita pkg2 promotoru konstantní v průběhu celé růstové křivky a přímá analýza RNA prokázala přítomnost transkriptu pkg2. Analýza transkripčního počátku pomocí S1 nukleázového mapování prokázala, že pkg2 mRNA postrádá vedoucí sekvenci. Během hladovění na fosfát byla promotorová aktivita pkg2 detekovatelná pouze v průběhu časnéexponenciální fáze. Snížení aktivity pkg2 promotoru a úbytek transkriptu se časově překrývalo se začátkem produkce granaticinu
