    Three new chromatographical forms of Dichomitus squalens MnP were isolated from wheat-straw cultures using MonoQ and CIM fast protein liquid chromatography. Enzymes revealed identical molecular masses of 50 kDa (estimated by SDS-PAGE) and pI values of 3.50, however, they varied in Km values obtained for Mn2+ oxidation. The addition of wood and straw methanol extracts to D. squalens cultures showed that the production of MnPs in wheat-straw fungal cultures was influenced rather by the type of cultivationthan by phenolic compounds from lignocellulosic material which induced laccase production. The purified CIM1 MnP was able to decolorize selected azo and anthraquinone dyes more rapidly than D. squalens laccase Lc1 per unit of enzyme activity. In vitro dye decolorization also showed a synergistic cooperation of MnP and laccase. In CSB degradation MnP prevented from production of a differently colored product yielded from degradation by laccase/HBT system

    V této práci byly izolovány tři nové formy MnP z kultur houby Dichomitus squalens rostoucích na pšeničné slámě. Enzymy vykazovaly shodné molekulové hmotnosti a hodnoty pI= 3,5. Lišily se však v Km pro oxidaci Mn2+. Přidání metanolových extraktů slámy a borovicového dřeva k tekutým kulturám houby ukázalo, že produkce MnP houbou D. squalens je ovlivňována spíše typem kultivace než fenolickými látkami pocházejícími z lignocelulózových materiálů, které se ukázaly jako induktory produkce lakasy. PurifikovanáMnP byla testována na schopnost dekolorizovat syntetická azo a antrachinonová barviva. In vitro pokusy prokázaly, že enzym v tomto procesu synergisticky spolupracuje s lakasou. Během degradace CSB MnP byla zodpovědná za degradaci barevných produktů vznikajích oxidací barviva lakasou a mediatorem HBT
