Abstract.The cuticle forms an effective barrier protecting plants from water loss. On the other hand, foliar uptake of lipophilic, hydrophilic and ionic agrochemicals is intensively studied. Permeability for water and other compounds significantly differs between species, types of cuticles (stomatous, astomatous), and may be affected by environmental conditions. Enzymatic isolation of leaf cuticle allows obtaining intact cuticles for chemical composition and permeability measurements. Cuticular permeability to water is usually characterized by the variable permeance, which is commonly measured e.g. in stainless-steel transport chambers. This method is indirect and 3H2O labelled water is used for exact assessment of water permeability. We proposed a new method based on an on-line detection of water flux from liquid phase to atmosphere through isolated leaf cuticles in semi-flow chambers. This approach is new in using the phenomenon of surface plasmon resonance for detection of liquid phase refractive index affected by water vapor. Isolated cuticles of hypostomatic leaves of Ficus elastica and Hedera helix were used for method evaluation. Permeances of stomatous and astomatous leaf cuticles were calculated under different flow rates of air and compared with values obtained by weight measurements of water permeability through isolated cuticles mounted in stainless-steel chambers and with literature data.
