    Autocrine VEGF Signalling in Malignant Astrocytromas

    Malignant astrocytomas are among the most angiogenic tumours. Both, pro- and anti-angiogenic mechanism control tumour neovascularization. The observation that angiogenic inhibitors decrease tumour growth in vivo suggests that angiogenesis is not merely an epiphenomenon of malignancy. Because diffusely infiltrating topic of angiogenesis and its relation to tumour progression and resistance to therapy is of special interest for novel research approaches. Exuberant angiogenesis is a critical event in astrocytoma progression and the key pro-angiogenic factor VEGF is often found over-expressed in high grade astrocytomas.

    Malígne astrocytómy predstavujú skupinu výrazne vaskularizovaných nádorov. Nádororová neovaskulararizácia je kontrolovaná pro- aj proti-angiogénnymi mechanizmami. Keďže angiogénne inhibítory znižujú rast nádoru in vivo, predpokladá sa, že angiogenéza nieje len akýmsi sprievodným javom malignity. Difúzne infiltrujúce astrocytómy sú neúspešne liečené operatívnymi a adjuvantnými prístupmi, preto je problematika angiogenézy a jej vzťahu k nádorovej progresii a rezistencii voči terapii doležitým objektom záujmu súčasného výskumu. Masívna angiogenéza je kritickým dejom v pregresii astrocytómov, pričom kľúčový proagniogénny faktor ? VRGF je často nadmerne exprimovaný u malígnych variant nádorov mozgu.
