The development of biosensor for the determination of amylase activity

a-Amylase (EC 3.2.1.1) catalyze the hydrolysis of starch to a significant extent resulting in the release of maltose. Determination of serum a-amylase activity is a procedure widely used, for example, in clinical medicine for the diagnosis of acute pancreatitis. The procedures most frequently used to determine a-amylase activity are either amyloclastic or saccharogenic (MAZZUCHIN et al., 1973). The former is based on the starch-iodine reaction and the latter on the formation of reducing sugars, measuredas maltose or glucose equivalents. Biosensor determination of maltose is based on an oxygen Clark type electrode equipped with a suitable enzymatic membrane (FILIPIAK et al.,1996). This membrane can be obtained by immobilization of the enzymes hydrolysing maltose (a-glucosidase) to glucose, which is determined using an oxygen electrode with the glucose oxidase membrane.
