Application of Q-TOF technology in plant hormone research
Original language description
Combination of high performance liquid chromatography (HPLC) or capillary liquid chromatography (CapLC) with quadrupole-time of flight mass spectrometry (Q-TOF) makes possible a new direction in plant hormone research. The instrument facilities are a good alternative for search of new plant metabolites and metabolic pathways. The recently developed method for quantitative analysis of natural cytokinins (Novák et al., 2003) was adapted from single-quadrupole mass spectrometer (SQ-MS) to Q-TOF micro instrument. The full scan spectra at very low concentration levels (100 fmol) were obtained together with MS-MS analyses of selected ions. The accurate masses of the parent ions and their fragments were calculated and used for determination of elementary structure with fidelity 10 ppm and better with a external calibration.
