The importance of cytokinins during Rhodococcus fascians pathology has been established over 40 years ago In Arabidopsis thaliana, the cytokinin perception machinery and subsequent signal transduction proved to be highly induced upon infection. Via mutant analysis we unequivocally show that the AHK3 and AHK4 cytokinin receptors are crucial for symptom development, underlining the central role for cytokinins in disease. Cytokinin profiling of the supernatant of R. fascians cultures allowed the identification of three classical cytokinins (iP, tZ and cZ) and their 2MeS-derivatives (2MeSiP, 2MeStZ and 2MeScZ). Although these six cytokinins are detected both in the pathogenic strain D188 and in its non-pathogenic derivative D188-5, the levels of all, except 2MeSiP, suggest that they originate from de novo synthesis encoded by the linear plasmid, and are therefore associated with virulence. Unexpectedly, the 2MeS-cytokinins are not cytotoxic at high concentrations.
