Detection of reactive oxygen species based on chemiluminescence of luminol and Cypridina Luciferin Analogues
Original language description
In connection with the mechanism of bioluminescence of Cypridina luciferin/ Cypridina luciferase system, the chemiluminescence of various Cypridina luciferin analogues is intensively investigated. 2-Methyl-6-phenyl-3,7-dihydroimidazol/1,2-a/pyrazin-3-one(CLA) or 2-methyl-6-(methoxyphenyl)-3,7-dihydroimidazol/1,2-a/pyrazin-3-one (MCLA) react with O2.- or 1O2 to emit light. The luminol-H2O2-CuSO4, CLAs-H2O2-CuSO4, luminol-H2O2-HRP, CLAs-H2O2-HRP, luminol-NaOCl-H2O2 and CLAs-NaOCl-H2O2 systems were studied at 25°C and 37°C. Superoxide dismutase (SOD, a scavenger of O2.-) and NaN3 (a quencher of 1O2) were used for differentiation between O2.- and 1O2-dependent luminescence.
