We examined the role of caspase proteases in carbonyl cyanide m-chlorophenyl-hydrazone (mCCCP) -induced apoptosis of human promyelocytic HL-60 cells. Treatment of HL-60 cells with micromolar concentration of mCCCP resulted in cell death due to mitochondrial failure of ATP synthesis. Cell death induced by mCCCP exhibited typical apoptotic features such as chromatin condensation, apoptotic bodies formation, nucleosomal fragmentation of DNA, and distinct increase in caspase 3 activity. However, although mCCCP-induced apoptosis was accompanied by activation of caspase 3, its full inhibition by selective inhibitor, Z-DEVD-FMK, neither prevented cell death
