Original language description
We worked out the method of analysis of polyphenol substances for optimization of splitting the mixture of polyphenol substances and also for determination of ferulic acid in various kinds of Czech beers with HPLC technique with gradient elution using electrochemical detector Coulochem II and photo diode array detector. The SPE technique using methanol, water and acetonitrile as solvents was employed for sample treatment. To optimize polyphenol mixture splitting mobile phases with sodium phosphate and ammonium acetate were used. The value of optimal pH in both mobile phases was identified at 3.5. The mobile phase with sodium phosphate was only used for quantitative determination of ferulic acid in respective kinds of beers due to a better signal response on detectors. The measured values of ferulic acid ranged from 2.48 mg.l-1 to 10.84 mg.l-1, which corresponds with the literature data.
