Shoot regeneration from various explants of horse chestnut (Aesculus hippocastanum L.)

  • Original language description

A protocol is described for the adventitious shoot bud regeneration from leaf, stem and root explants in A. hippocastanum. Different medium (Heller, WPM, Rugini and DKW) and growth regulators [N6-benzyladenine (BA), thidiazuron (TDZ), and naphthaleneacetic acid (NAA)] at a range of various concentrations have been investigated for organogenesis. Nutrient content in the culture media had a significant effect on regeneration of new shoots in Aesculus. The highest number and length of shoots was achieved on Rugini and WPM medium with moderate nutrient content. Adventitious shoot formation was observed in all types of explants. Regeneration frequency and shoot number depended on explant type and growth regulators. The highest regeneration (100%) and shootbud number (9.68) were recorded in stem explants on medium with 2 mg l--1 BA + 0.1 mg l--1 NAA. Generally, TDZ has lower effect on shoot regeneration in stem and leaf explants compared to BA. The additions of NAA to the culture medium imp
