    Optimization of Metafectene-mediated transfection of metallothionein down-regulating siRNA in triple-negative breast cancer cells

  • Original language description

    Small interfering RNA (siRNA) has become an important instrument for studying gene functions. These double strand RNA oligonucleotides are characterized by targeted mRNA degradation after introduction of specific sequence RNA. Efficiency of siRNA is the basis of gene silencing and gene therapy. In this case, we optimized siRNA transfection to achieve down regulation of gene expression. Then, we validated this optimization on specific sub isoform of metallothionein, MT-1X, into triple-negative breast carcinoma cell lines. In this project, we focused on achieving results with the best possible targeted gene suppression with high viability of cells and high transfection effectivity.
