Original language description
The systematic health control of sweet chestnut (Castanea sativa Mill.) population in the Czech Republic is carrying out from 2001. More than 1,000 samples collected from 282 localities were checked. Presence of Cryphonectria parasitica (Murrill) M. E. Barr, the causal agent of the chestnut blight, was confirmed in three localities in Moravia. In two localities, Uherský Brod (locality nr. 1) and Kuřim (locality nr. 2), only individual chestnut trees were afflicted, in the third locality in Moravský Písek dozens of trees in an ornamental tree nursery were infected. Twenty pure virulent cultures of C. parasitica were isolated from 195 bark samples in locality nr. 3. Two of them were from Quercus rubra. Samples of the bark were washed in sterile water, surface-sterilised (96% ethanol 1 min, sterile water, dipping in ethanol), and placed in Petri dishes containing 3% malt extract agar (MA3).
