Neurodegenerative diseases, which encompass a broad spectrum of debilitating conditions, are among the most incapacitating ailments affecting humans. These diseases are characterized by the progressive degeneration of neurons in the brain, leading to impairments in cognition and mobility. Alzheimer's disease (AD) is one of the most prevalent neurodegenerative disorders, with over 30 million confirmed cases worldwide. The initial symptom of AD is often repeated episodes of short-term memory impairment, which gradually worsens and impacts more complex mental capabilities, including speech and reasoning abilities. Coumarin derivatives have emerged as promising candidates for AD treatment due to their diverse chemical structures and wide range of pharmacological properties. In addition to their acetylcholinesterase inhibition activity, these compounds exhibit several other biological effects that are beneficial in addressing various aspects of AD pathology. For instance, coumarin derivatives can inhibit secretase-1, a key enzyme involved in the production of amyloid beta plaques, a hallmark of AD. Additionally, they can act as monoamine oxidase inhibitors, modulate gamma-aminobutyric acid (GABA) receptors, and inhibit cyclooxygenase and lipoxygenase enzymes, which are involved in inflammation and oxidative stress. Furthermore, coumarin derivatives also act as blockers of cannabinoid receptor 2 and antagonists of the NMDA receptor, which play roles in synaptic dysfunction and neurodegeneration. Given these multifaceted biological effects, coumarin-based compounds show significant potential for the treatment of AD. This chapter delves into the therapeutic significance of coumarin derivatives in AD, exploring their mechanisms of action and their potential to address the underlying causes of the disease, such as neuroinflammation, oxidative stress, and amyloid plaque formation.
