Original language description
To evaluate the impact of the stress induced by thermotherapy on the primary structure of DNA, AFLP (Amplified Fragment Length Polymorphism) was applied in this study. Epigenetic changes were determined by means of MSAP (Methylation Sensitive Amplified Polymorphism). Studies were carried out on plants at different times after thermotherapy treatments (6 weeks to 5 years after it). The results show nearly identical AFLP-derived fingerprints in all analysed samples from plants treated by in vitro thermotherapy, suggesting that the stress condition did not provoke changes at the sequence level or the method is not suitable to detect possible changes. In contrast, MSAP allowed the observation of differences in epigenetic fingerprints in thermotherapy treated plants. Furthermore, the method allowed to determine the impact in a time dependent and quantifiable fashion.
