    The presence of cytokinins in the culture medium and their active form in explants is decisive for organogenesis in vitro. The accumulation of cytokinins and multiple shoot formation on leaf segment of tobacco and petunia was studied using an addition ofBAP (6-benzylamino-purine) or iP (isopentenyladenine) and/or mT (meta-topoline) in concentration of 1.5 mg/l to the induction medium. The aim of this study was to compare the cytokinin activity in morphogenesis. The leaf explants of petunia formed multiple shoots after 9 days of culture on medium containing BAP or mT more intensively than tobacco leaf explants, while they regenerated early after 5 days on a medium with iP. A higher content of endogenous cytokinins (riboside) and taken up cytokinins (BAP, iP) was detected after 5 days (tobacco) and 9 days (petunia) of culture than the long-term culture of explants.
